- Capacity: Growth area of 6.15 square inches.
- Shape: Square design.
- Color: Transparent.
- Dimensions: 3″ x 3″ x 4″.
Supreme Tissue-Culture-Grade Agar
- Description: A polysaccharide derived from the red alga Rhodophyceae, processed via bleaching and hot water extraction. Recommended usage rate: 6–12 g/L. Supreme agar ensures excellent clarity for culture media.
- Gel Strength: 1050 GM/CM².
- Packaging Options: 100g, 500g, 1kg in poly bottles.
- Applications: Ideal for solid microbial culturing media, it’s used at concentrations of 0.5–1.0% (w/v) to solidify plant tissue culture media. Essential for in vitro root development.
Murashige & Skoog (MS) Medium
- Description: MS medium with vitamins and calcium chloride (without sucrose and agar).
- Technical Details: Supports plant growth and shoot proliferation across both monocot and dicot tissue cultures.
- Formula Weight: 4.54 g/L.
Preparation Instructions:
-
- Dissolve 4.54 g of powder in 600 ml of distilled or deionized water at room temperature (15–30°C).
- Rinse the media vial with a small amount of distilled water to remove residue.
- Add desired heat-stable supplements before autoclaving.
- Stir until dissolved. If needed, lower pH to ~3.0 for better dissolution.
- Adjust pH between 5.4 and 5.8 using 1N HCl/NaOH/KOH.
- Top up to 1 liter with distilled water. Mix gently and heat until clear. Avoid boiling or reheating.
- Autoclave at 121°C (15 psi) for 15 minutes.
- After cooling to 45–50°C, add heat-sensitive supplements aseptically.
- Storage: Keep at 2–8°C, away from moisture and direct light. Use before the expiry date. Opened containers should be used immediately or stored in desiccators at 2–8°C.
Plant Preservative Mixture (PPMâ„¢)
- Description: PPMâ„¢ is a heat-stable preservative and biocide designed to prevent or minimize microbial contamination in plant tissue culture.
- Key Benefits:
- Effectively inhibits bacteria and fungi spore germination.
- Maintains in vitro seed germination, callus proliferation, and regeneration.
- Heat-stable and autoclavable with culture media.
- More cost-effective than traditional antibiotics.
- Reduces contamination from airborne, waterborne, and human contact sources.
- Can be used to mitigate endogenous contamination in some cases.
- Usage: PPMâ„¢ is ideal as a standard ingredient in tissue culture media and protects against contamination even with rigorous sterile practices.
